細(xì)胞因子檢測(cè)
細(xì)胞因子檢測(cè)有助于了解其在免疫調(diào)節(jié)中作用, 鑒定分離淋巴細(xì)胞, 監(jiān)測(cè)某
些疾病狀態(tài)細(xì)胞免疫功能。例如, 根據(jù)培養(yǎng) CD4+T 細(xì)胞分泌細(xì)胞因子確定細(xì)胞
亞群, 產(chǎn)生 IL - 12 和 IFN -γ者為 Th1 細(xì)胞, 產(chǎn)生 IL - 4 和 IL - 10 者為 Th2 細(xì)胞; 艾滋
病患者 IL - 2 水平明顯降低, 而類風(fēng)濕性關(guān)節(jié)炎、多發(fā)性硬化、移植排斥反應(yīng)等患者
IL - 2水平則升高。細(xì)胞因子檢測(cè)方法可分以下 3 種:
(1) ELISA。用細(xì)胞因子單克隆抗體包被固相雙抗體夾心法或 ELISPOT 測(cè)定法檢
測(cè)細(xì)胞因子, 例如檢測(cè) IL - 2, IL - 5, IL - 6。亦可用熒光素標(biāo)記細(xì)胞因子抗體, 對(duì)胞
內(nèi)細(xì)胞因子進(jìn)行染色, 直接以流式細(xì)胞儀測(cè)定產(chǎn)生該細(xì)胞因子細(xì)胞數(shù)目。
(2) 生物活性測(cè)定法。可根據(jù)細(xì)胞因子生物活性, 選用相應(yīng)實(shí)驗(yàn)系統(tǒng), 包括細(xì)
胞增殖法、直接殺傷法、保護(hù)細(xì)胞免受病毒致病變法等。如選用細(xì)胞因子依賴細(xì)胞
株, 其增殖反應(yīng)與細(xì)胞因子量成正相關(guān), 根據(jù)細(xì)胞株增殖水平可確定樣品中細(xì)胞因
子含量, 如檢測(cè) IL - 1, IL - 2, IL - 4, IL - 6。
(3) 聚合酶鏈反應(yīng) (PCR)。根據(jù)細(xì)胞因子核苷酸序列, 設(shè)計(jì)特定細(xì)胞因子引
物, 利用逆轉(zhuǎn)錄 PCR 測(cè)定待檢細(xì)胞中特異 mRNA。該法可用于多種細(xì)胞因子測(cè)定。 |
